SPOP(speckle-type POZ protein), is a substrate adaptor of a cullin3-RING ubiquitin ligase (CRL3) that recruits substrates to the ligase for ubiquitination and subsequent proteasomal degradation. SPOP typically localizes to nuclear speckles and has also been reported to localize to DNA damage loci, promyelocytic leukemia (PML) bodies, and other substrate-containing bodies. SPOP contains a substrate-binding MATH domain and two dimerization domains, BTB and BACK. SPOP self-associates into large higher-order oligomers through the synergistic function of two dimerization domains, the BTB and the BACK domains.
